摘要: The effect of recombinant tumor necrosis factor-alpha (rTNF alpha) on human natural killer (NK) function was examined. Lysis both the NK-sensitive K562 erythroleukemia line and relatively insensitive renal carcinoma Cur by nonadherent peripheral blood lymphocytes significantly enhanced as a result an 18-hr preincubation with either rTNF alpha or interleukin 2 (rIL 2). When cells were preincubated low doses rIL (1 to 10 U/ml), marked additional augmentation lysis targets noted which greater than that caused cytokine alone. Similar results observed when responses CD16+ large granular selected fluorescence-activated cell sorter after staining NK-specific monoclonal antibody Leu-11 examined, indicating action cytokines directly cytotoxic cells. Augmentation could not be ascribed cytolytic activity targets, because no combination 2, alpha, interferon-gamma Cur. By flow cytometric analysis, it found expression IL receptors induced purified alone even degree 2. Additional analysis surface antigens blocking studies antibodies showed leukocyte function-associated antigen-1-mediated effector/target interactions. These data indicate alone, in augments NK activity.
