摘要: If present at all, endogenous DNA within an ancient sample will have experienced two general damage processes, analogous to those seen in vivo. The quantity of potential PCR template and the size amplifiable fragment be limited by cross-linking, radiation-induced double-strand breaks, hydrolytic oxidative depurination, formation hydantoins. In addition this, even when amplification is successful, sequence likely contain miscoding lesions as a result further damage. While these processes are hindrance aDNA researcher, they also provided us with new insights into significance secondary structure on vivo mitochondrial mutation, hence evolution. As studies postmortem degradation undertaken, it can hoped that help resolve both hotly contested debate heteroplasmy, presence mutational hotspots.
