摘要: The α- and β-globin gene clusters have been extensively studied1,2,3. Regulation of these genes ensures that proteins derived from both loci are produced in balanced amounts, expression is tissue-restricted specific to developmental stages. Here we compare the subnuclear location endogenous primary human cells which either actively expressed or silent. In erythroblasts, localized areas nucleus discrete α-satellite-rich constitutive heterochromatin. However, cycling lymphocytes, do not express globin genes, distribution was markedly different. loci, common with several inactive studied here (human c-fms SOX-1) previously (mouse λ5, CD4, CD8α, RAGs, TdT Sox-1)4,5, were associated pericentric heterochromatin a high proportion lymphocytes. contrast, α-globin centromeric normal lymphocytes patients α-thalassaemia lacking regulatory HS-40 element entire upstream region locus, mouse erythroblasts transgenic mice. These data show regulated occurs different nuclear environments haemopoietic cells.
