摘要: Abstract Background: Interleukin 1 (IL-1) is a key mediator of bowel inflammation, but there limited knowledge about the amount and site production this cytokine in gastrointestinal tract under physiological or pathological conditions. Methods: Epithelial lamina propria mononuclear cells were isolated from control, Crohn's disease- ulcerative colitis-involved mucosa to investigate capacity these generate IL-1 bioactivity, IL-1α IL-1β immunoreactivity, gene expression. Results: Control produced substantial amounts IL-1β, which increased dramatically when inflammatory disease used. disease, colitis intestine displayed no bioactivity immunoreactivity. Lamina contained moderate large quantities messenger RNA (mRNA), respectively, whereas epithelial had none. The absence transcripts was selective, because mRNA for HLA-DR antigens present control cells. Conclusions: In normal inflamed human distinct compartmentalization IL-1, as not cytokine. high levels may explain several its local systemic manifestations, blockade by specific antagonists could have important therapeutic effects.
