In vitro ageing of pig oocytes: effects of the histone deacetylase inhibitor trichostatin A.

摘要: After in vitro maturation, the unfertilized pig oocytes underwent process called ageing. This involves typical events such as fragmentation, spontaneous parthenogenetic activation or lysis. Inhibition of histone deacetylase, using its specific inhibitor trichostatin A (TSA), significantly delayed maturation cultured vitro. The ageing matured under effect TSA is same without TSA. inhibition deacetylase during oocyte reduced percentage fragmented (from 30% untreated to 9% aged 100 nM TSA). Oocytes and subsequently for 1 day effects retained their developmental capacity. activation, a higher portion (27% vs. 15%) developed blastocyst stage after 24 h when compared with activated TSA-free medium. development treatment similar fresh (29% blastocyst) artificially immediately maturation.
