Cytochrome P450 in the central nervous system as a therapeutic target in neurodegenerative diseases

摘要: Cytochromes P450 (CYPs) constitute a family of enzymes that can be found in the endoplasmic reticulum (ER), mitochondria or cell surface cells. CYPs are characterized by carrying out oxidation organic compounds and they mainly recognized as mediators biotransformation xenobiotics to polar hydrophilic metabolites eliminated from organism. However, these play key role many other physiological processes, being involved diverse indispensable metabolic pathways since metabolize endogenous substrates. Various CYP isoforms expressed brain, it is believed this could part due particular function brain CYPs. In cholesterol turnover, biosynthesis dopamine, serotonin, morphine, hormones, protective lipid (epoxyeicosatrienoic acids), addition their already detoxification psychotropic drug metabolism. Increasing evidence suggests group fundamental for normal functioning maintenance homeostasis. This review focused on highlighting importance CYP-mediated metabolism central nervous system (CNS) its relationship with recent findings regarding involvement neurodegenerative diseases. Some therapeutic approaches regulation also discussed.
