Notch-mediated conversion of activated T cells into stem cell memory-like T cells for adoptive immunotherapy

摘要: Adoptive T-cell immunotherapy is a promising approach to cancer therapy. Stem cell memory T (TSCM) cells have been proposed as class of long-lived and highly proliferative cells. CD8+ TSCM can be generated in vitro from naive via Wnt signalling; however, methods do not yet exist for inducing activated or Here, we show strategy generating TSCM-like (iTSCM cells) CD4+ mice humans by coculturing with stromal that express Notch ligand. iTSCM lose PD-1 CTLA-4 expression, produce large number tumour-specific effector after restimulation. This method could therefore used generate antigen-specific adoptive immunotherapy.
