Increased expression of transgene in stably transformed cells of Dunaliella salina by matrix attachment regions

摘要: Nuclear matrix attachment regions (MARs) are known to bind specifically the nuclear scaffold and thought influence expression of transgenes. In our previous studies, a new deoxyribonucleic acid fragment isolated from Dunaliella salina could in vitro had typical characteristics MARs. this study, investigate effects MARs on transgenes stably transformed cells D. salina, vectors with without MARs, which contained chloramphenicol acetyltransferase (CAT) reporter gene driven by ribulose 1,5-bisphosphate carboxylase/oxygenase promoter, were constructed delivered, respectively, into electroporation. Twenty colonies randomly picked out, CAT was assayed. The results showed that enzyme vector containing averaged out about 4.5-fold higher than those while transgene variation among individuals transformants decreased threefold. lines not significantly proportional copy numbers, suggesting may be through increasing numbers.
