摘要: The kinetochore is the macromolecular protein complex that assembles onto centromeric DNA and binds spindle microtubules. Evolutionarily divergent kinetoplastids have an unconventional set of proteins. It remains unknown how kinetochores assemble at centromeres in these organisms. Here, we characterize KKT2 KKT3 kinetoplastid parasite Trypanosoma brucei. In addition to N-terminal kinase domain C-terminal polo boxes, proteins a central function. We show are important for localization several their domains sufficient centromere localization. Crystal structures from two reveal unique zinc-binding (termed CL localization), which promotes its T. Mutations equivalent abolish Our work shows play critical role mediating KKT3.
