Matrix metalloproteinases activity in fibrosarcoma Wehi-164 cells

摘要: Fibrosarcoma is a malignant disorder of fibroblasts and extremely metastatic cancer. Angiogenesis plays an important role in metastasis. Several factors including matrix metalloproteinases (MMPs) involve angiogenesis. MMPs are large group endopeptidases which degredate the extracellular matrix. The present study was conducted to evaluate patterns MMP-2/MMP-9 activity Wehi-164 cells in vitro. cells were cultured complete RPMI-1640 medium. Then seeded at density 4 ×105 cells/ml andwere incubated with phorbol myristate acetate (PMA) (25 ng/ml) or phytohaemagglutinin (PHA) (10 µg/ml) for 24, 48 72 h. cell-conditioned media then evaluated by gelatin zymography. PHA/PMA significantly increased MMP-2 time-dependent manner. MMP-9 shown unstimulated time-dependently. PHA did not show any significant effect on whereas PMA induced Wehi-164, time dependently. According result study, Wehi 164 fibrosarcoma could potentially exhibit activity. However, activity, varies according presence absence stimulator as well duration incubation time. Thus, cell line be useful screening tool modulators.    Key words: Fibrosarcoma, lines, metalloproteinase-2 (MMP-2), metalloproteinase-9 (MMP-9).
