摘要: Abstract Antibiotic concentrations associated with antibiotic bone cements may cause skeletal cell toxicity and prevent fracture healing. We investigated effects of dose treatment time after exposure to three antibiotics commonly used in orthopaedic local drug delivery systems. hypothesized a threshold exists for osteoblasts chondrocytes ciprofloxacin, vancomycin, or tobramycin. To test this hypothesis, we first determined whether caused differences cellular morphology. Cells exposed ciprofloxacin showed considerable changes spread, membrane, extensions. next asked what dosage would reductions osteoblast chondrocyte numbers. Ciprofloxacin at greater than 100 microg/mL vancomycin tobramycin doses 2000 severely decreased proliferation. Finally, questioned observed decreases numbers were the result increased senescence. Released lactate dehydrogenase ratios osteoblasts. These data suggest balance between targeted microbicidal host is critical survival function.
