Biochemical Evidence That Small Proline-rich Proteins and Trichohyalin Function in Epithelia by Modulation of the Biomechanical Properties of Their Cornified Cell Envelopes

摘要: The cornified cell envelope (CE) is a specialized structure involved in barrier function stratified squamous epithelia, and assembled by transglutaminase cross-linking of several proteins. Murine forestomach epithelium undergoes particularly rigorous mechanical trauma, these CEs contain the highest known content small proline-rich proteins (SPRs). Sequencing analyses revealed that SPRs as cross-bridgers joining other use multiple adjacent glutamines lysines on only amino carboxyl termini functionally non-polar ways. Forestomach also trichohyalin novel cross-bridging protein. We performed mathematical modeling acid compositions mouse human epidermis different body sites. Although sum loricrin + was conserved, amount varied relation to presumed physical requirements tissues. Our data suggest could serve modifiers composite CE material composed mostly loricrin; we propose increasing amounts modify CE, just strengthen types In this way, epithelia may varying alter biomechanical properties tissue accordance with specific functions.
