摘要: Abstract Qβ replicase (RNA-directed RNA polymerase of bacteriophage Qβ) exponentially amplifies certain RNAs in vitro . Previous studies have shown that can initiate and elongate on a variety RNAs; however, only minute fraction them are recognized as ‘legitimate’ templates. Guanosine 5′-triphosphate (GTP)-dependent initiation legitimate template generates stable replicative complex capable elongation the presence aurintricarboxylic acid, powerful inhibitor RNA–protein interactions. On contrary, an illegitimate is GTP independent does not result aurintricarboxylic-acid-resistant complex. This article demonstrates 3′ 5′ termini cooperate during after step. Breach cooperation by dividing into fragments or introducing point mutations at terminus reduces rate yield initiation, increases requirement, decreases overall copying, destabilizes postinitiation These results revive old idea functional circularity templates complement increasing body evidence may be common property directing synthesis either protein molecules.
