摘要: 2.1. Lighting for a total reflection fluorescence measurement has previously been done by means of prism on the side facing away from microscope objective, wherein sample to be investigated must prepared with great effort prism. TIRF illumination alternatively takes place through requiring high numeric aperture, and thus complex due large angle incidence required. The invention relates having axial resolution at low complexity. 2.2. A lighting device (1) is designed as module comprises an optical fiber (2) collimating optic (3), mounted in front light discharge opening fiber, such that it collimates exiting divergently into bundle, excitation can applied outside detection beam path. numerical aperture decoupled detection, standard objective sufficient detection. 2.3. Fluorescent microscopy.
