摘要: The present invention discloses a method for amplifying hematopoietic stem cells based on 3D culture system. comprises: (1) placing cell suspension separated from blood into composite medium to culture, wherein the comprises serum-free base medium, SIFE growth factor, serum alternate and fibrin gel; (2) in step hypoxia incubator, maintained oxygen concentration incubator is 3-20%. has following advantages that: gel added polymer network system, can absorb lot of water, structure similar extracellular matrix, simulate vivo microenvironment so as provide sufficient spaces space structures supporting communications between cells, reduce cost, significantly increase amplification multiple maintain characteristics prevent loss due differentiation.
