摘要: Regulation of actin dynamics at filament ends determines the organization and turnover cytoskeletal structures. In striated muscle, it is believed that tight capping fast-growing (barbed) by CapZ slow-growing (pointed) tropomodulin (Tmod) stabilizes uniform lengths (thin) filaments in myofibrils. Here we demonstrate for first time both Tmod are dynamic on basis rapid incorporation microinjected rhodamine-labelled (rho-actin) barbed pointed from photobleaching green fluorescent protein (GFP)-labelled Tmod. Unexpectedly, inhibition GFP-Tmod overexpression results shorter thin filaments, whereas cytochalasin D has no effect length. These data myofibrils relatively despite presence proteins, regulated assembly length filaments.
