Cell-cycle dependent anti-FGF-2 staining of chicken cardiac myocytes: Movement from chromosomal to cleavage furrow- and midbody-associated sites

摘要: Fibroblast growth factor-2 (FGF-2) promotes cardiac myocyte proliferation and has been detected in extracellular as well cytoplasmic nuclear compartments. As a first step examining the participation of intracellular FGF-2 cell cycle we have investigated its localization proliferative chicken cells during interphase various stages mitosis culture. We used previously characterized affinity-purified anti-FGF-2 antibody preparation which recognizes 19-22 kDa variants chick FGF-2. By immunofluorescence, bright, punctate labelling was observed 26% nuclei from myocytes derived 5 day embryonic heart ventricles; these were positive for anti-bromodeoxyuridine staining indicating that they are at S- or G2 phase cycle. In prophase metaphase, bright apparent association with chromosomes. During anaphase, however, dissociated chromosomal locations distinctly remaining strand-like structures area ensuing cleavage furrow formation. late telophase cytokinesis, strong persisted midbody reappeared small fraction newly formed daughter nuclei. Absorption immobilized eliminated all staining. This dynamic pattern suggests immunologically related protein(s) not only increase DNA-synthesizing but may play role subsequent cytokinesis.
