摘要: Costimulation via the B7/CD28 pathway is an important signal for activation of T cells. Maximal inhibition T-cell and induction alloantigen-specific nonresponsiveness in vitro was achieved using anti-CD80 monoclonal antibody (mAb) combination with cyclosporin A (CsA). Based on this knowledge, efficacy prophylactic treatment mAb CsA allogeneic skin graft survival tested a preclinical rhesus monkey model. No side effects have been observed. Administration resulted high serum levels that decreased to undetectable values around day 7. At same time, anti-mouse response started develop. The bound peripheral blood mononuclear cells detectable lymph node grafted during period. time untreated or suboptimally CsA-treated monkeys 10 days. Treatment (blood 100-160 ng/ml) (0.5 mg/kg) significantly increased 14 Eventually, grafts all were rejected, which coincided increase helper cytotoxic frequency directed against donor antigens.
