Methods of preparing low-toxicity drug-lipid complexes

摘要: Methods and compositions are described for nonliposomal lipid complexes in association with toxic hydrophobic drugs such as the polyene antibiotic amphotericin B. Lipid preferably a combination of phospholipids dimyristoylphosphatidylcholine (DMPC) dimyristoylphosphatidylglycerol (DMPG) about 7:3 mole ratio. The contain bioactive agent, may be made by number procedures, at high drug:lipid ratios. These (HDLCs) administered to mammals humans treatment infections, substantially equivalent or greater efficacy reduced drug toxicities compared their free form. Also disclosed is novel liposome-loading procedure, which also used formation HDLCs.
