The effects of experimental conditions of fluorescence quenching on the binding parameters of apigenin to bovine serum albumin by response surface methods

摘要: A fluorescence quenching technique is often used to study interactions between small molecules and serum albumin. However, the results are quite different by using spectroscopic techniques on same drug-protein interaction research they may be affected conditions (e.g. working solution of pH ionic strength). In this research, apigenin as an example, effect experimental binding parameters drug bovine albumin was investigated a response surface method (RSM). The pH, concentration NaCl Mg(2+) constant (KSV), apparent association (Ka) number sites (n) studied single-factor experiments with [NaCl] [Mg(2+)] independent variables KSV, Ka n values. Prediction models were fit quadratic polynomial regression equation showed that both KSV displayed second-order model, whereas linear relation dependence [Mg(2+)]. Under these conditions, most significant (p < 0.05) impact factor Ka, 0.05).
