摘要: Inhalation of irritants, such as toluene diisocyanate (TDI), stimulates substance P (SP) release from peripheral processes sensory neurons innervating the airways. The purpose this study was to determine if TDI inhalation affects intraneuronal levels SP and preprotachykinin (PPT) messenger RNA (mRNA) in trigeminal ganglion (TG) which innervate nasal epithelium. cavity Fisher-344 rats instilled with rhodamine-labeled latex microspheres. Ten days later, were exposed 60 ppb 2,4-2,6-TDI vapor for 2 h. TG removed 1, 12, 24, 48, 72, 96 h after treatment prepared immunocytochemistry PPT situ hybridization. nerve fiber density epithelium significantly increased 48 exposure. proportion microsphere-labeled cell bodies expressing high immunoreactivity decreased at 24 but above controls 72 mRNA control percentage leukocytes observed lavage fluid inhalation. These studies indicate that production projecting is transiently exposure, suggesting not only causes also neuropeptide levels. Increased neuronal may be involved maintaining neurogenic inflammation or development airway hyperresponsiveness.
