摘要: Purpose:Aberrant activation of the hedgehog (Hh) pathway is implicated in pancreatic ductal adenocarcinoma (PDAC) tumorigenesis. We investigated prognostic and predictive value four Hh signaling proteins tumor stromal density. Experimental Design:Using tissue microarray immunohistochemistry, expression Shh, Gli1, SMO PTCH1were assessed 567 patients from three independent cohorts who underwent surgical resection for PDAC. In 82 patients, index (SI) was calculated its association with overall survival (OS) disease-free (DFS) investigated. Results:Shh Gli1 protein abundance were factors resected PDACs; low expressors those experiencing a better OS DFS. The combination Shh levels most significant predictor defined 3 clinically relevant subgroups different prognosis (Gli1 low; HR set at 1 v 3.08 or high 5.69 high; P< 0.001). two validating recapitulated findings training cohort. After further stratification by lymph node status, significance combined maintained. SI correlated significantly associated (p = 0.023). Conclusions:Shh are biomarkers
