摘要: Background and objective: COPD is a global disease characterized by chronic bronchitis obstructive emphysema. Its pathogenesis not fully understood. This study aimed to use proteomics provide new insights into the mechanisms of COPD. Methods: Protein lysates were prepared from lung tissue samples harvested never-smokers, non-COPD smokers smokers, analysed using 2-dimensional gel electrophoresis. Differentially expressed proteins identified mass spectrometry. The differential expression heat shock protein 27 (Hsp27) cyclophilin A (CyPA) was validated immunohistochemistry western blotting. Results: Twenty-four spectrometry as being differentially among three groups subjects. main functions these involve basic metabolism, oxidation/reduction, coagulation/fibrinolysis, degradation, signal transduction, inflammation cell growth/differentiation/apoptosis. Proteomic analysis revealed that Hsp27 CyPA upregulated in this upregulation particularly marked smokers. variation between confirmed blotting. Conclusions: are associated with COPD, smoking contributes overexpression proteins.
