Rapid Plant Regeneration and Molecular Assessment of Genetic Stability Using ISSR and RAPD Markers in Commercial Banana Cv. Grand Naine (G-9)

摘要: The investigation was carried out to assess the genetic stability in   tissue culture raised plants of banana cv. G-9 using random amplified polymorphic DNA (RAPD) and inter simple sequence repeats (ISSR) markers. Aims: Molecular assessment tissue molecular Material Results: Apical shoots were established on medium EM 4  (MS + BAP 4.0 mg L -1 ) with maximum 3.8 buds/explant 2.6 days. bud multiplication with 16.5±0.06 shoots was observed medium Ma 3 medium+ 5.0  BAP 0.25  NAA of + 30  AdSO ). The rooting response (100%) was observed 1/2 MS medium supplemented 2.0 -1  NAA in 12.2 After acclimatization hardened examined for RAPD ISSR primers. Total forty six (twenty twenty ISSR) markers used. primers produced 87 distinct scorable bands, an average 3.34 bands per primer amplification products range from 100-1200 bps. number varied 2 5 primer. 71 100-1000 bps each 6 3.55 Conclusion: Similar profile monomorphic all when compared mother plant both types results corroborate fact that technology has immense importance production true type planting material.
