摘要: A simple and sensitive HPLC-fluorescence assay was developed for the determination of a gastroprokinetic agent mosapride in small volumes rat plasma. Samples (50 μL) were treated with 200 μL internal standard solution (cisapride, 0.1 μg/mL acetonitrile). Chromatographic separation achieved on C18 column by gradient elution mobile phase acetonitrile-water containing 20 mM potassium dihydrogen phosphate, at flow rate 1 mL/min. Fluorescence measured excitation emission set 315 354 nm, respectively. The retention time about 16 min cisapride mosapride. No endogenous substances found to interfere. calibration curve linear from 0.015 10 μg/mL. lower limit quantification intra- inter-day precision expressed as relative deviation did not exceed 7.7%, accuracy within 4.7% nominal concentration. method used successfully investigate disposition kinetics rats.
