摘要: In response to various environmental signals, macrophages (MΦ) acquire distinct functional phenotypes that have been classified as classical MΦ and alternatively activated (AAM). AAM cells sustain tissue repair, whereas their depletion contributes aberrant wound healing. The current study was aimed at determining whether polarization participated in epithelium repair lung fibrosis regulation. To this aim, we used the vivo model of injury induced by intra-peritoneal injection naphthalene mice. Epithelium phase accompanied an increased expression bronchoalveolar lavage (BAL) cells, mRNAs encoding markers (CD206, FIZZ-1 YM1), AAM-derived mediators, MMP-12, CCL2 elastin. FACS analysis on BAL demonstrated a significant increase number monocytes during proceeded complete (days 6 15). To define role depleted administering intra-nasally liposome clodronate naphthalene-injected mice days 2, 5 8. MΦ-depleted displayed impaired excessive production fibrogenic mediators (IL-6 CCL2) lower levels TIMP-1 lung, enhanced collagen deposition 21 35, compared control mice. Intra-tracheal adoptive transfer purified day11 after administration restored suppressed fibrosis. We conclude is towards phenotype, essential for driving preventing fibrosis.
