摘要: A truncated version of the nef gene simian immunodeficiency virus SIVmac239 capable encoding amino acids 98 to 263 was used as bait screen a cDNA library from activated lymphocytes in yeast two-hybrid system. The zeta chain T-cell receptor (TCRzeta) found interact specifically not only with SIV cells but also full-length glutathione S-transferase (GST)-SIVnef fusion protein vitro. Coimmunoprecipitation TCRzeta demonstrated transfected Jurkat and Cos 18 which express cytoplasmic domain fused external CD8 via transmembrane domain. Using series deletion mutants, we have mapped binding site within central core (amino 235). Binding specific for isolated SIVmac239, SIVsmH4, human (HIV)-2ST detected five different HIV-1 isolates. An active tyrosine kinase coprecipitated nef-TCRzeta complexes J.CAM1.6 lack functional Lck kinase. These results demonstrate association HIV-2 nef, TCRzeta.
