Investigation of TEMPO partitioning in different skin models as measured by EPR spectroscopy - Insight into the stratum corneum

摘要: Electron paramagnetic resonance (EPR) spectroscopy represents an established tool to study properties of microenvironments, e.g. investigate the structure and dynamics biological artificial membranes. In this study, partitioning spin probe 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) in ex vivo human abdominal breast skin, porcine ear skin as well normal inflammatory vitro equivalents was investigated by EPR spectroscopy. Furthermore, stratum corneum (SC) lipid composition (as determined high-performance thin-layer chromatography), SC chain order (probed infrared spectroscopy) thickness (investigated histology) were models. X-band measurements have shown that TEMPO partitions lipophilic hydrophilic microenvironment varying ratios different Ex exhibited highest amount microenvironment. contrast, lowest equivalents. Individual spectra epidermis including dermis indicated mainly corresponds most part epidermis, SC. The independent but correlated with thickness. conclusion, could be a novel technique determine differences thickness, thus suitably complementing existing methods.
