摘要: Hippocampal slices were prepared under three conditions: (1) in medium containing glucose and oxygen at 4 degrees C; (2) as (1), but 37 (3) devoid of C. The rates recovery to roughly steady-state levels through 8 h incubation monitored for energy metabolite related parameters. In vitro stable values are compared with situ hippocampal levels. Regardless the conditions which prepared, required up 3 stabilize, these maintained or improved incubation. Further, maximal concentrations metabolites independent slice preparation. Total adenylates total creatine reached 55% those vivo. Lactate decreased from decapitation-induced high levels, stabilized about twice rapidly frozen brain. Cyclic AMP cyclic GMP exhibited peak 30 min incubation, remained elevated h. Although all methods preparation resulted similar profiles on initial decreases phosphates delayed by chilling. Most striking, absence much smaller orthodromic evoked potentials dentate gyrus. presence during appears be critical electrophysiological response tissue.
