摘要: Abstract We report mutations affecting several steps of ColE1 primer formation. Primer precursor forms the hybrid with template DNA and is cleaved by RNAase H to form primer. Point at positions −264, −265, −268, −308 (base pairs upstream replication origin) reduce efficiency A suppressor mutation −18 increases for a mutant transcript reduces that wild-type transcript. Therefore, formation probably starts after transcription passes this position. −10 affects site cleavage H. The appears be determined distance from stem-loop structure immediately upstream. double −186 −188 results in an RNA secondary prevents use H-cleaved as
