摘要: DNA topoisomerase II (topo II) is an essential nuclear enzyme required for chromatin condensation and chromosome segregation during mitosis. Forced overexpression of topo IIα was found to cause morphological changes in recipient cells associated with apoptosis. This induction apoptosis localization IIα, yet independent the cleavage-religation activity enzyme. Apoptosis mediated by deregulation blocked crmA, a specific inhibitor certain caspases, but not bcl-2. IIα-induced also dominant-acting mutant stress-activated protein kinase (SEK1/MKK4) its normal counterpart. Furthermore, coexpression dominant-negative form cyclin-dependent cdk2 cdc2. These results provide rationale tight regulation levels through cell cycle that expression apoptotic death.
