摘要: Abstract Wolfiporia cocos Ryvarden et Gilbertson, a well-known medicinal fungus in the Basidiomycetes, is widely distributed East Asia. Its dried sclerotium, which known as Fuling China, has been used traditional crude drug Chinese medicine for thousand years. However, little about how sclerotium developed at genetic level. In this study, de novo sequencing of sclerotia W. (S1_initial stage; S2_developmental stage and S3_mature stage) was carried out by illumina HiSeq 2000 technology. 27,438 unigenes were assembled from ~ 30 Gbp raw data, 12,093 significantly annotated. The analysis expression profiles during development returned 304 differentially expressed genes (DEGs), clustered into four different groups according to their trends. Especially maturation (S3), exhibited markedly profile other stages. We further showed that peroxisome, unsaturation fatty acids degradation pathway respectively prevalent S1, S2 S3 stages evidenced enrichment analysis. To our knowledge, study represents first report sclerotial transcriptomics cocos. obtained results provide novel insights developmental biology sclerotia, helpful future studies cultivation breeding
