摘要: A human malignant pleural mesothelioma cell line (STAV) was studied with respect to production of the extracellular matrix components laminin, type IV collagen, and fibronectin, interactions these proteins in vitro. We also analyzed STAV serum-free conditioned medium possible presence "autocrine motility factor-like" substance. Sodium dodecylsulfate-polyacrylamide gel electrophoresis biosynthetically labeled showed that cells released several into medium, including molecular weights 850,000, 540,000 440,000. Using Western blotting we identified as respectively. By immunocytochemistry fibronectin were detected a surrounding cells. Plastic culture dishes coated microgram quantities induced attachment spreading Laminin, stimulated directional (chemotactic) migration Boyden chambers fitted 8 microns filters. The same migrated insoluble step gradients filter-bound (haptotaxis). When separated by using fast protein liquid chromatography Superose 6 filtration, two motility-inducing peaks detected. first peak contained weight > 220,000 had both chemotactic haptotactic properties, while second material apparent approximately 67,000 chemokinetic (random motility) but not properties. Analysis M(r) indicated it heat-sensitive trypsin-digestible protein. soluble motile response molecules well autocrine factor-like substance may be important for highly invasive behavior this tumor.
