摘要: Hepatitis B virus core protein virus-like particles (HBc-VLPs) act as a strong immunogen and are suitable for uptake by dendritic cells (DCs), in which they directly promote DC maturation migration. To illustrate the utility of global proteomic analysis techniques elucidating molecular events that altered HBc-VLP-pulsed bone marrow-derived DCs (BMDCs) to gain better understanding mechanisms capture processing BMDCs, an antigen (Ag) delivery system based on BMDCs was developed. Two-dimensional electrophoresis (2-DE) tandem mass spectrometry (MS/MS) analyses were utilized analyze differential expression patterns between untreated BMDCs. Protein spots with significantly levels detected, identified validated. The results showed exogenous HBc-VLPs phagocytosed efficiently enhanced efficacy BMDC Ag presentation, VLPs also induced high Ag-specific CD8+ T displayed cytotoxic lymphocyte (CTL) activity vivo. Several differentially expressed proteins, including growth factor receptor bound 2 (Grb2) annexin A2 (AnxA2), detected analysis, validated western blot.
