Genetic response to metabolic fluctuations: correlation between central carbon metabolism and DNA replication in Escherichia coli

摘要: Until now, the direct link between central carbon metabolism and DNA replication has been demonstrated only in Bacillus. subtilis. Therefore, we asked if this is a specific phenomenon, characteristic for bacterium perhaps its close relatives, or more general biological rule. We found that temperature-sensitivity of mutants particular genes coding proteins could be suppressed by deletions certain enzymes metabolism. Namely, effects dnaA46(ts) mutation dysfunction pta ackA, dnaB(ts) pgi pta, dnaE486(ts) tktB, dnaG(ts) gpmA, dnaN159(ts) ackA. The observed suppression were not caused decrease bacterial growth rate. genetic correlation exists model Gram-negative bacterium, E. coli. This at steps initiation elongation replication, indicating important global metabolic status cell events leading to reproduction.
