摘要: The nematode Caenorhabditis elegans is characterized by many features that make it highly attractive to study nuclear pore complexes (NPCs) and nucleocytoplasmic transport. NPC composition structure are conserved in nematodes being amenable a variety of genetic manipulations, key aspects envelope dynamics can be observed great details during breakdown, reassembly, interphase. In this chapter, we provide an overview some the most relevant modern techniques allow researchers unfamiliar with C. embark on studies nucleoporins intact organism through its development from zygote aging adult. We focus methods generate loss-of-function phenotypes their analysis advanced microscopy.
