Synthesis of atrial natriuretic polypeptide in human failing hearts. Evidence for altered processing of atrial natriuretic polypeptide precursor and augmented synthesis of beta-human ANP.

摘要: To elucidate the synthesis of atrial natriuretic polypeptide (ANP) in failing heart, 20 human right auricles obtained at cardiovascular surgery were studied. The concentration alpha-human ANP-like immunoreactivity (alpha-hANP-LI) ranged from 13.8 to 593.5 micrograms/g, and tissue alpha-hANP-LI severe congestive heart failure (CHF) (New York Heart Association [NYHA] functional class III IV) (235.4 +/- 57.2 micrograms/g) was much higher than that mild CHF (NYHA I II) (52.5 15.6 micrograms/g). Atrial levels significantly correlated with plasma concentrations these patients (r = 0.84, P less 0.01). High performance gel permeation chromatography reverse phase high liquid coupled radioimmunoassay for ANP revealed auricle consisted three major components ANP, gamma-human (gamma-hANP), beta-human (beta-hANP) (alpha-hANP). Comparing percentages gamma-hANP, beta-hANP, alpha-hANP those CHF, predominant component gamma-hANP whereas beta-hANP and/or prevailing and, especially, markedly increased hearts. These results demonstrate total atrium is increase mainly due small molecular weight forms alpha-hANP, especially indicate processing precursor, or differs normal suggesting augments secretion as one its own compensatory responses.
