Intracellular adenosine 5′-triphosphate, adenosine 5′-diphosphate, and adenosine 5′-monophosphate detection by short-end injection capillary electrophoresis using methylcellulose as the effective electroosmostic flow suppressor

摘要: We present a new rapid CE method to measure adenine nucleotides adenosine 5'-triphosphate (ATP), 5'-diphosphate (ADP), and 5'-monophosphate (AMP) in cells. The short-end injection mode allows decrease the analysis time by injecting samples at outlet end of silica capillary closest detection window, reducing migration distance. Moreover, use methylcellulose (MC) as run buffer additive suppress EOF permits further reduce times analytes. Thus, when with an effective length 10.2 cm was used 60 mmol/L sodium acetate pH 3.80 presence 0.01% MC, analytes were 1.35 min for ATP, 1.85 ADP, 4.64 AMP. These conditions gave good reproducibility intra- interassay (CV <4 8%, respectively) all procedure demonstrated excellent analytical recovery (from 98.3 99 %). suitability proved both on red blood cells spermatozoa. compared our proposed spectrophotometric assay, measuring ATP levels 40 spermatozoa samples. obtained data analyzed Passing Bablok regression Bland-Altman test.
