Effect of silodosin on detrusor overactivity in the male spontaneously hypertensive rat

摘要: What's known on the subject? and What does study add? Recently, several studies have suggested that detrusor overactivity (DO) is result of bladder ischaemia. Hypertension could affect pelvic arterial blood flow, resulting in loss smooth muscle with resultant compliance. Spontaneously hypertensive rats are considered a valuable tool for exploring pathogenesis DO. Some reports indicate α1 adrenoceptor antagonists improve chronic ischaemia lower urinary tract patients LUTS, concomitant improvement their symptoms as well DO through an increased flow (BBF) rat outlet obstruction. However, mechanism silodosin storage or irritative not investigated still unclear. Silodosin prevents hypertension-related SHR via possible mechanisms. One efficacy to includes BBF. OBJECTIVE •  To investigate effect α1A selective antagonist, silodosin, its spontaneously (SHRs). MATERIALS AND METHODS •  Twelve-week-old male SHRs received treatment (100 µg/kg perorally) once daily 6 weeks; vehicle-treated Wistar were used our study. •  Six weeks after treatment, voiding functions estimated by behaviour cystometric all groups. •  The was measured hydrogen clearance method, tissue levels nerve growth factor (NGF) calcitonin gene-related peptide (CGRP) enzyme-linked immunosorbent assay (ELISA). •  Furthermore, expressions subtype mRNAs real-time PCR method. RESULTS •  The showed significant increases pressure, micturition frequency NGF CGRP bladder. •  Moreover, decreases single voided volume both compared those rats. •  Six significantly ameliorated alterations these variables small changes pressure. •  The expression similar groups rank order =α1D > α1B groups. •  However, there no differences between any groups. CONCLUSION •  The data present suggest normalizes SHRs, which be related bladder.
