摘要: ATP-binding cassette (ABC) transporters constitute a large superfamily of proteins that mediate transport diverse number substrates including nutrients, lipids and xenobiotics across membranes serving variety developmental physiological functions. Here, we report on the molecular properties biological roles ABC transporter LmABCH-9C in migratory locust Locusta migratoria. was expressed continuously during nymphal development all tissues integument. Expression highest just after molting. Suppression transcript levels by RNA interference (RNAi) nymphs provoked death or soon molting to next stage. These lost weight within minutes Moreover, high humidity rescued lethality molted dsLmABCH-9C-injected nymphs. In histological experiments, find amounts inner-cuticular are reduced with suppressed expression. data together indicate is needed for lipid-dependent desiccation resistance, paralleling function ABCH-9C Tribolium castaneum. Hence, this seems be conserved insect species ranging from hemimetabolous (L. migratoria) holometabolous (T. castaneum) species. addition, cuticle inward impermeability compromised function. summary, consistent model cuticular necessary prevent penetration insects, hypothesize involved construction lipid-based barrier at surface especially protect animal against uncontrolled water loss entry. Susceptibility RNAi-mediated knockdown designates it as an excellent target RNAi-based pest control.
