Epithelial to Mesenchymal Transition Might be Induced via CD44 Isoform Switching in Colorectal Cancer

摘要: Background Isoform switching of CD44 is associated with epithelial to mesenchymal transition (EMT) in several cancers; however, the clinical implications this remain unclear for colorectal cancer (CRC). Methods We measured expression levels E-cadherin, vimentin, standard (CD44s) and variant 9 (CD44v9) transcripts 14 CRC cell lines 150 patients. We determined EMT status by calculating vimentin/E-cadherin CD44s/CD44v9 ratios, respectively. Associations between isoform switching, clinicopathological factors prognosis were analyzed. Results CD44s was highly expressed mesenchymal-type lines, while CD44v9 epithelial-type lines. knockdown resulted decreased vimentin expression, significantly reduced proliferation, migration invasion cells. In patients, group high exhibited poorer survival than that (5-year survival; 62.1% vs. 85.5%, P = 0.0085) low 66.1% 85.0%, P = 0.0251). On multivariate analysis, an independent prognostic factor.
