摘要: The advances in the molecular neurosciences during “the decade of brain” conjunction with new genomic and proteomic sciences have radically changed paradigm central nervous system (CNS) drug design development. classical, chemistry-based small-molecule discovery has been broadened by twenty-first century biology-driven large-molecule neuropharmaceuticals: peptides, recombinant proteins, monoclonal antibodies, antisense oligonucleotides, gene therapy. As this chapter will discuss, however, despite progress field neuroproteomics rational strategies, there is still a high failure rate candidates later stages projects consequently an unsatisfied demand for novel neurotherapeutic neurodiagnostic medicines. Therefore, CNS targeting must be merged early development process order to obtain neuropharmaceuticals active brain following systemic administration.
