摘要: Abstract The objective of this work was to develop a simple surface modification technique using keratin derived from human hair for efficient cardiomyogenic lineage commitment mesenchymal stem cells (hMSCs). Keratin extracted discarded containing both the acidic and basic components along with heterodimers. adsorbed conventional tissue culture polystyrene surfaces at different concentration. solution 500 μg/ml yielded well coated layer 12 ± 1 nm thickness minimal agglomeration. promoted cell attachment proliferation. Large increases in mRNA expression known cardiomyocyte genes such as cardiac actinin, troponin β-myosin heavy chain were observed. Immunostaining revealed increased sarcomeric α-actinin tropomyosin whereas Western blots confirmed higher myocyte enhancer factor 2C on than non-coated surface. DNA demethylation Atp2a2 Nkx2.5 thereby elucidating importance epigenetic changes possible molecular mechanism underlying differentiation. A global gene analysis significant alteration involved pathways associated including cytokine chemokine signaling, cell–cell cell-matrix interactions, Wnt MAPK TGF-β signaling FGF among others. Thus, adsorption offers facile affordable yet potent route inducing important implications developing xeno-free strategies cardiovascular regenerative medicine.
