Quantifying viable Vibrio parahaemolyticus and Listeria monocytogenes simultaneously in raw shrimp

摘要: A novel TaqMan-based multiplex real-time PCR method combined with propidium monoazide (PMA) treatment was firstly developed for the simultaneous quantification of viable Vibrio parahaemolyticus and Listeria monocytogenes in raw shrimp. The optimization PMA concentration showed that 100 μM considered optimal to effectively inhibit 108 CFU/mL dead cells both bacteria. high specificity this confirmed on tests using 96 target non-target strains. optimized assay could detect as low 101–102 CFU/g each strain artificially contaminated shrimp, its amplification efficiencies were up 106 % V. L. monocytogenes, respectively. Furthermore, has been successfully applied describe behavior these two pathogens shrimps stored at 4 °C. In conclusion, technique, where whole procedure takes less than 5 h, provides an effective rapid tool monitoring contamination seafood, improving seafood safety protecting public health.
