Increase of mitochondria surrounding spindle causes mouse oocytes arrested at metaphase I stage.

摘要: During oocyte meiosis, mitochondria usually surround spindle to meet the energy demand of migration and chromosome segregation. Therefore, mitochondrion surrounding is widely accepted as an important indicator demonstrate mitochondrial function in studies. However, role quality not exactly addressed. Mitofusin-2 (MFN2) a outer membrane GTPase that mediates clustering fusion. Here, we increased by overexpression MFN2 mouse oocytes. Results indicate increase has little effect on germinal vesicle breakdown (GVBD), migration, potential (MMP), reactive oxygen species (ROS) production Endoplasmic reticulum (ER) distribution, while blocks segregation, destroys spindle, finally causes most oocytes arrest at metaphase I stage. Collectively, our results precisely regulated during maturation, too much it may cause abnormal meiosis. although typical biological event utilizing should be careful.
