摘要: Light triggers chloroplast differentiation whereby the etioplast transforms into a photosynthesizing and thylakoid rapidly emerges. However, sequence of events during remains poorly understood. Using Serial Block Face Scanning Electron Microscopy (SBF-SEM), we generated series 3D reconstructions differentiation, revealing number volume extent envelope membrane surfaces. Furthermore, used quantitative lipid whole proteome data to complement (ultra)structural data, providing time-resolved, multi-dimensional description differentiation. This showed two distinct phases biogenesis: an initial photosynthesis-enabling 'Structure Establishment Phase' followed by 'Chloroplast Proliferation cell expansion. Moreover, these detail expansion de-etiolation at seedling level relative contribution differential regulation proteins lipids each developmental stage. Altogether, establish roadmap for critical process plant photoautotrophic growth survival.
