摘要: Differentiated rat thyroid epithelial cells, infected in vitro with a temperature-sensitive mutant of the Kirsten murine sarcoma virus, expressed at permissive temperature (33 degrees C) some phenotypic properties typical transformed including morphological features, colony formation agar, and induction tumors newborn animals. Specific functional markers these differentiated i.e., synthesis/secretion thyroglobulin, synthesis thyroglobulin mRNA iodide uptake, were blocked during growth 33 C. Normal morphology, failure to grow requirement hormones for optimal all restored after shifting nonpermissive transformation (39 C), though functions remained blocked. Infection leukemia helper virus clone (Moloney or virus) did not lead loss phenotype thus demonstrating that is caused by component. These results indicate expression cells under direct control p21 thermosensitive activity, whereas block two differentiation irreversible probably controlled different mechanisms.
