Sequence-based genetic markers for genes and gene families: single-strand conformational polymorphisms for the fatty acid synthesis genes of Cuphea

摘要: Gene sequences are rapidly accumulating for many commercially and scientifically important plants. These resources create the basis developing sequence-based markers mapping tracking known (candidate) genes, thereby increasing utility of genetic maps. Members most gene families underlying synthesis seed oil fatty acids have been cloned from medium-chain oilseed Cuphea. Allele-specific-PCR (AS-PCR) single-strand conformational polymorphism (SSCP) were developed 22 acid genes belonging to seven Cuphea using homologous heterologous DNA sequences. Markers 4 fatty-acyl-acyl carrier protein thioesterase, 2 β-ketoacyl-acyl synthase I, II, 3 III, acyl protein, reductase, enoyl-acyl reductase loci. Eighty-eight percent (14 16) SSCP loci polymorphic, whereas only 9% (2 22) AS-PCR polymorphic. mapped a viscosissima×C. lanceolata F2 population produced linkage groups 10, 3, (3 segregated independently). The 10-locus group had every but one necessary 2- 16-carbon acetyl-CoA malonyl-ACP (the missing family was not mapped). analysis has broad fingerprinting families.
