摘要: The FHIT (fragile histidine triad) gene located at chromosome 3p14.2 has been proposed as a candidate tumor suppressor in human cancers. Fhit protein with the diadenosine 5',5'”-P1,P3-triphosphate (Ap3A) hydrolase activity is product of gene. way which exerts its and relationship Ap3A to suppression are not known. As step toward understanding function cell we have explored intracellular localization distribution rat tissues. Data obtained from immunoblot analysis showed that was most abundant spleen brain. Moderate amount detected kidney liver, whereas level heart, skeletal muscle glomeruli undetectable. RT-PCR performed on RNA isolated these tissues no product, mRNA spleen, brain, kidney, liver lung correlated level. subcellular fractions various by differential density-gradient centrifugation localized exclusively nucleus plasma membrane. Presented data showing nuclear membrane may support hypothesis concerning signaling molecule.
