Characterization of an immunomodulatory Der p 2-FIP-fve fusion protein produced in transformed rice suspension cell culture.

摘要: Der p 2, a major allergen of Dermatophagoides pteronyssinus mites, is one the most clinically relevant allergens to allergic patients worldwide. FIP-fve protein (Fve) from golden needle mushroom (Flammulina velutipes) an immunomodulatory with potential Th1-skewed adjuvant properties. Here, we produced and immunologically evaluated 2-Fve fusion as immunotherapeutic for diseases. Using inducible expression system in cultured rice suspension cells, recombinant (designated OsDp2Fve) was expressed cells under control α-amylase gene (αAmy8) promoter secreted sucrose starvation. OsDp2Fve partially purified medium. The conformation 2 remains intact reflected by its unaltered allergenicity, assessed human IgE ELISA histamine release assays, compared non-fusion protein. Furthermore, Fve retains vitro lymphoproliferative activity but loses hemagglutination lymphoagglutination effects native Notably, vivo evaluation showed that mice administered possessed enhanced production 2-specific IgG antibodies without potentiating Th2 effector cytokines comparison co-administered proteins.
